MoveTip #1: Skyline Quick Start: Extracted Ion Chromatograms
⚡ Brief Summary It is often difficult to know “where to start” with Skyline. An easy place to start is to use it to perform extracted ion chromatograms for molecules of interest from a raw data file…but it seems intimidating! In this video, we demonstrate how to jump-start your Skyline journey by performing extracted ion chromatograms from LC or CE-MS data in small molecule mode. This one focuses on high resolution! 🧪 What You’ll Learn • How to set up a Skyline document to import raw data • How to add molecules to the Skyline document • How to import the raw data for visualization of the extracted ion chromatograms 💻 Try It Yourself Here is how you can test this yourself right away: • Make a list of your molecules of interest in excel, including a molecule name, molecular formula, and ionization adduct (ie [M+H], [M-H], etc) • Settings / Transition settings to set the full scan parameters • Ctrl+C (Copy) in excel to copy your molecule list including molecule name, molecular formula, and ionization adduct • Ctrl+V (Paste) into Skyline to assign the molecules you want to measure.
