The story takes place during the turbulent late Meiji period. Saichi Sugimoto, a hero of the Russo-Japanese War, earned the nickname "Invincible Sugimoto" for his demonic fighting style, including at Hill 203, the most brutal battlefield of the Russo-Japanese War. He spends his days panning for gold in Hokkaido, hoping to obtain a large sum of money for a specific purpose. There, Sugimoto learns of the existence of a huge amount of gold that had been stolen from the Ainu people. The man who stole the gold, Nopperabo, hid it in a certain location just before being captured, and tattooed the location on the bodies of 24 prisoners, allowing them to escape. The tattoos of all 24 prisoners form a single code. Just then, Sugimoto is attacked by a wild brown bear, but is saved by an Ainu girl. The girl, named Asirpa, had her father killed by the man who stole the gold. Sugimoto, who is pursuing the gold, and Asirpa, who wants to avenge her father, end up working together. Also aiming for the gold is Lieutenant Tsurumi Atsushirou of the Imperial Japanese Army's 7th Division. For the sake of the division members who risked their lives in the Russo-Japanese War but were not rewarded, Golden Kamuy is plotting to conquer Hokkaido and needs the gold as military funds. Another escapee is Shinsengumi's "Demon Vice-Commander," Hijikata Toshizo, who was thought to have died in the Boshin War.
