DEG-77 CK1α degrader targets TP53 WT gynecologic cancers with MDM4 dependence and SWI/SNF alteration

This talk explores CK1α as an emerging therapeutic target in oncology, and how next-generation molecular glue degraders such as DEG-77 can uncover vulnerabilities in TP53 wild-type gynecologic cancers. In SWI/SNF-deficient ovarian and endometrial cancer models, CK1α degradation disrupts MDM4-mediated suppression of p53, restoring p53 activity to drive apoptosis and cell cycle arrest, resulting in strong antiproliferative effects in these challenging cancer types.
