If the puncture is in the front wheel, you could always put full lock on and manoeuvre the car to provide a good position for you to repair the tyre. If the puncture is in the rear wheel, you will most likely need to take it off to be able to apply enough force to penetrate the tyre with the reamer tool. Remember these kits are not sold as permanent repairs for cars, they are designed as a temporary repair until you can take the wheel to a tyre shop to get it professionally patched or replaced.
