How does it interact? Speakers: Stephen McLaughlin, Chris Batters, Tomas Morgan, and Trevor Rutherford MRC Laboratory of Molecular Biology, UK In the first part of this video, Stephen discusses the issues of reproductivity of data and unconscious bias. He describes issues that must be considered to make the best measurements. He then gives an overview of biophysical techniques which can be used to analyse protein complex formation. These include isothermal titration calorimetry (ITC), analytical ultracentrifugation (AUC), fluorescence polarisation (FP), microscale thermophoresis (MST), biosensors. Chris continues with an overview of single molecule techniques including optical tweezers. Tomas, then demonstrates the various strategies to map interactions with mass spectrometry including Bio-ID, crosslinking and hydrogen-deuterium exchange (HX). In the last part of the video, Trevor shows the multiple ways NMR can be used to probe biomolecular interactions giving site specific information. The LMB Biophysics Facility houses a wide range of state-of-the-art and in-house built instruments that enable the molecular characterisation of macromolecular size, shape, mass, stability, binding affinity, dynamics and interaction kinetics. The facility is primarily intended for hands-on research by students, postdocs, and group leaders, who prepare samples and make measurements. Biophysics supports users in advice on experimental techniques and results, training and in collaborative research. The LMB Mass Spectrometry and Proteomics Facility houses a wide range of mass spectrometers enabling biological proteomics and structural proteomics. Biological proteomics involves the identification of proteins and their interactions and the comparative proteomics of cells and tissues. In structural proteomics purified proteins or their complexes are investigated by hydrogen-deuterium exchange or cross-linking mass spectrometry. The LMB NMR Facility contributes to projects across the full range of research activities at the LMB and is part of an integrated structural biology approach, bridging the gap between biophysical measurements and structural determination. It is a biology driven facility that makes NMR accessible to the non-specialist user. Facility staff draw on their diverse scientific experience so that all relevant advanced NMR methods can be implemented and adapted to the specific needs of the LMB scientists.
