Sterilization refers to any process that removes, kills, or deactivates all forms of life (in particular referring to microorganisms such as fungi, bacteria, spores, unicellular eukaryotic organisms such as Plasmodium, etc.) and other biological agents like prions present in a specific surface, object or fluid, for example, food or biological culture media. Wet Heat/Steam Sterilization- In most labs, this is a widely used method that is done in autoclaves. Autoclaves use steam heated to 121–134 °C under pressure. Autoclaving kills microbes by hydrolysis and coagulation of cellular proteins, which is efficiently achieved by intense heat in the presence of water. The intense heat comes from the steam. Pressurized steam has high latent heat and at 100°C it holds 7 times more heat than water at the same temperature. In general, Autoclaves can be compared with a typical pressure cooker used for cooking except in the trait that almost all the air is removed from the autoclave before the heating process starts. Wet heat sterilization techniques also include boiling and pasteurization. At constant temperatures, sterilization times vary depending on the type of item (e.g., metal versus rubber, plastic, items with lumens), whether the item is wrapped or unwrapped, and the sterilizer type. Mode of Action = Moist heat destroys microorganisms by the irreversible coagulation and denaturation of enzymes and structural proteins. In support of this fact, it has been found that the presence of moisture significantly affects the coagulation temperature of proteins and the temperature at which microorganisms are destroyed. An autoclave is a machine that provides a physical method of sterilization by killing bacteria, viruses, and even spores present in the material put inside of the vessel using steam under pressure. Autoclave sterilizes the materials by heating them up to a particular temperature for a specific period of time. An autoclave is used to sterilize surgical equipment, laboratory instruments, pharmaceutical items, and other materials. It can sterilize solids, liquids, hollows, and instruments of various shapes and sizes. Autoclaves vary in size, shape and functionality. A very basic autoclave is similar to a pressure cooker; both use the power of steam to kill bacteria, spores and germs resistant to boiling water and powerful detergents.
