Quadruplet Versus Triplet Therapy for Multiple Myeloma: Dr. Jesús San-Miguel Shares Key Trial Data

Jesús San-Miguel, MD, PhD, of Clínica Universidad de Navarra, presented efficacy and safety data from the PERSEUS and CEPHEUS trials, which demonstrated that daratumumab plus bortezomib, lenalidomide, and dexamethasone (D-VRd) improved measurable residual disease negativity (MRD) and progression-free survival (PFS) compared to VRd. “Quadruplets have become the newest standard of care both for the transplant and the non-transplant patients,” he began. “They were associated with deeper responses as compared with triplets and prolonged PFS. We have no yet data for overall survival, but it’s expected.” The estimated progression-free survival for D-VRd in the PERSEUS trial is 17 years, Dr. San-Miguel noted. Despite a slight increase in infections with quadruplet therapy, the safety profile is generally comparable between quadruplet and triplet therapy.
