This is the process of glycogen synthesis, an animal storage polysaccharide made up of glucose monomers linked linearly by alpha (1-4) glycosidic bonds and with abundant branches formed by alpha (1-6) glycosidic bonds. The key enzyme in this process is glycogen synthase, which is responsible for joining glucose molecules from a non-reducing end of a pre-existing glycogen branch. Although it is a very efficient enzyme with an enormous catalytic rate, it has three important limitations: 1. It cannot add glucose directly; instead, the glucose must be "activated" in the form of UDP-glucose (a precursor of glycogen). 2. It requires a primer. If there is no glycogen branch to which to add the UDP-glucose, it cannot act. The enzyme glycogenin is responsible for creating new glycogen nuclei when necessary. 3. It does not branch the glycogen chain. It cannot form α(1-6) linkages, so the branching enzyme (amyl(1-4,1-6) transglucosylase) will form the branch, which can later be elongated by glycogen synthase.
