The right (Sure)TAC for membrane protein removal | LATSS 2026
Speaker: Richard Sainson Synopsis: Membrane proteins are key drivers of diseases such as cancer and autoimmune disorders. While several approved therapies aim to block these proteins, selective degradation offers clear advantages, including deeper pathway inhibition, improved tissue and cell selectivity, and the ability to target previously “undruggable” proteins. Laigo is developing SureTAC™ (SUrface REmoval TArgeting Chimeras), a novel technology that harnesses the cell’s endo-lysosomal system to degrade membrane proteins. SureTACs are heterobifunctional molecules that bring a target into proximity with a transmembrane E3 ligase, triggering ubiquitination, endocytosis, and lysosomal degradation of the target. Using PD-L1 as a proof-of-concept, we demonstrate that SureTACs can effectively remove the PD-L1 from the surface of cancer cells in vitro and in vivo. Laigo is advancing oncology and I&I therapies by partnering with LifeArc to generate antibodies using the B-SMArT mouse platform, combining SureTAC™ and LifeArc’s discovery capabilities to accelerate the development of novel therapies and improve patient outcomes.
