Development of real-time imaging, spectral flow cytometry and strategically designed fluorochromes

Imaging flow cytometry has provided an additional layer of information for the characterization of cells based on the spatial distribution and localization of fluorescence signals. However, camera-based systems have so far been limited by the slow speed of image measurement, which prevented real-time cell sorting based on imaging parameters. In this presentation, we will describe how the BD CellView™ Image Technology was developed to overcome these limitations and enable, for the first time, high-speed image fluorescence cell sorting. We will furthermore describe how this technology was integrated in a 5-laser, 78 detector spectral flow cytometer. Lastly, we will introduce a new family of fluorochromes strategically designed to enable high-parameter flow and image cytometry and minimize the current challenges posed by the limited availability of unique, laser-specific fluorochromes.
