The municipal seat was discovered by Manoel Dias da Silva, at the beginning of the development of Goiás, around 1730, and is considered one of the first settlements in the State, founded simultaneously with the settlements of Barra and Meia Ponte. The possession of this area was symbolized by a Cross, with the inscription "Viva el-Rei de Portugal" (Long live the King of Portugal), justifying the name it received: Santa Cruz (Holy Cross). The first dwellings were formed next to the cross, and the construction of the Church of Nossa Senhora da Conceição marked the beginning of the formation of the settlement originating from mining, a preponderant factor for the development of the territory, creating an Intendency in 1733 for the collection of the royal fifth. It was originally inhabited by Indians, who were soon dispersed, enslaved blacks, adventurers and prospectors, and later by farmers and agricultural workers as a result of the decline in gold mining activity and the arrival of the railroad tracks. It is noteworthy in the history of Santa Cruz that it was for some time (there are no records of dates) the seat (Capital) of the Province of Goiás. On September 23, 1759, it became a parish and in 1809 it became a Judicial District. By Provincial Resolution of April 1, 1833, the settlement was elevated to the category of Town, established on December 8 of the same year. And by Law No. 735, of April 29, 1884, it obtained the status of City. In 1934, it lost its seat to Pires do Rio, and in 1943, it was renamed Corumbalina, until in 1947 it restored its status as a municipality and its previous name of Santa Cruz de Goiás.
