How close are we to using human engineered heart tissue (EHT) to diagnose cardiomyopathies and guide therapy? In this IEEE EMBS webinar, Prof. Stuart Campbell (Yale University) walks through a decade of research at the intersection of cardiac muscle physiology, multiscale computational modeling, tissue engineering, and clinical cardiology. He shows how iPSC-derived cardiomyocytes grown on decellularized myocardial scaffolds can capture human-relevant mechanics, illuminate genotype→phenotype pathways, and help classify variants of unknown significance (VUS). Using paired TPM1 mutations - E62Q (hypertrophic cardiomyopathy, HCM) and E54K (dilated cardiomyopathy, DCM) - the talk demonstrates opposite effects on force generation, length-dependent activation (Frank-Starling response), diastolic stiffness, and calcium sensitivity. Campbell also shares how mavacamten (myosin inhibitor) and danicamtiv (myosin activator) normalize contractile phenotypes in EHTs, and outlines a practical pipeline to functionally classify patient variants in weeks, plus early evidence that EHTs from “genotype-negative” HCM patients can still reveal contractile abnormalities.
