The question no one gets a full mark: Explain how mutation leads to a disease

Explain how the modification in the nucleotide sequence (mutation) leads to a disease. When a mutation in DNA occurs, transcription leads to a mutant mRNA that will be translated into a non functional proteins. This protein will not be able to make its function anymore, and therefore a disease will be manifested.
