Biological Mass Spectrometry and Proteomics - J. Mark Shekel
The LMB Mass Spectrometry and Proteomics Facility houses a wide range of mass spectrometers enabling biological proteomics and structural proteomics. Biological proteomics involves the identification of proteins and their interactions and the comparative proteomics of cells and tissues. In structural proteomics purified proteins or their complexes are investigated by hydrogen-deuterium exchange or cross-linking mass spectrometry. In this video Mark begins with a brief history of mass spectrometry and how samples are ionized and measured by the mass spectrometer. He then follows with a detailed explanation of mass-spectrometry approaches to biological proteomics via protein identification of single proteins and their interactions in cells via pull-down assay or proximity dependent biotin identification. In the last part of the talks the discusses different methods for quantitative proteomics via label-free, isotopic labelling or reporter ion-based quantitation. Finally he shows how post-translational modifications can be investigated. The Mass Spectrometry and Proteomics Facility is headed by J Mark Skehel.
