Why Your Compound Fails In Vivo - Sam Hoare
The binding affinity looked identical. Same target, same assay, same numbers on paper. One compound made it to Phase 2. The other did nothing in vivo. The difference came down to how long each molecule stayed on the CRF receptor — seven hours versus fifteen minutes — and no one had thought to measure it. Sam Hoare and the Neurocrin team had to build the framework to understand why their drug was working before they could build a better one. That instinct for the overlooked variable in pharmacology data has since become a career. About Sam Hoare Hoare is the founder of PharmaChanics, a pharmacology data analysis consultancy. After a PhD on allosteric modulation of dopamine receptors in Philip Strange's lab and a postdoc at the National Institutes of Health working on Class B GPCRs, Hoare spent 15 years at Neurocrin Biosciences advancing CRF receptor drug discovery programs. He now consults for biotech and pharma clients on GPCR data analysis, signaling kinetics, and pharmacological modeling. His current independent research focuses on developing accessible mathematical frameworks for analyzing kinetic GPCR signaling data — tools built specifically for use at the bench. What You'll Learn How a seven-hour receptor off-rate rescued a failing CRF drug discovery program — and how standard binding assays had missed it entirely Why signaling kinetics is the dimension most GPCR pharmacologists measure but almost none analyze with the same rigor they bring to dose-response data How Hoare developed Prism-compatible time-course equations and why he deliberately built them for early-career researchers first What biased agonism and receptor residence time actually mean as practical drug discovery tools, stripped of the hype cycles that surrounded them How to navigate the move from industry scientist to independent consultant — what you have to learn, what you discover about yourself, and what almost made Hoare give it all up Timestamps 00:00 Sponsor message 00:35 Introducing Sam Hoare: GPCR pharmacology data analyst 03:19 Origin story — the ATP calculation that sparked a scientific career 06:16 PhD with Philip Strange: allosteric modulation and the first aha moment 16:53 Why GPCRs?
