Centrifugation is the final step in the pre-analytical phase of clinical laboratory testing. It is the stage in which serum or plasma is obtained from the patient sample. Serum does not contain clotting factors, while plasma does; this is the main difference between the two sample types. Red, gold, or orange-top tubes are used to obtain serum, while blue, green, gray, or white-top tubes are used to obtain plasma. The choice of tube type depends on the intended application of the sample, as well as the specific resources and circumstances of each laboratory. Traditionally, revolutions per minute (RPM) have been used as the unit of centrifugation speed. However, the standard is to use g (grams per minute) so that the size of the centrifuge rotor does not affect the proper collection of serum or plasma. This video demonstrates how to convert between these units. Each manufacturer has its own recommendations regarding waiting time before spinning, spin speed, and spin time, and at the end of the video, these recommendations from at least three manufacturers are shown.
