In this video, we'll explain the Catalase Test in a simple and straightforward way. This is a very important and frequently asked practical test in microbiology, used to identify bacteria. This test is particularly useful for differentiating between bacteria like Staphylococcus and Streptococcus. (What is the Catalase Test?) The catalase test is a biochemical test that checks whether a bacterium produces the catalase enzyme. 👉 The main function of the catalase enzyme is to break down hydrogen peroxide (H₂O₂) to form water (H₂O) and oxygen (O₂). 👉 Reaction: 2H₂O₂ → 2H₂O + O₂ When oxygen gas is released, we see bubbles. These bubbles indicate the result of this test. ⸻ 🎯 Purpose of Catalase Test The main purpose of this test is: ✔ To differentiate catalase-positive and catalase-negative bacteria ✔ To aid clinical diagnosis ✔ To facilitate microbial identification ✔ To obtain rapid results in lab practicals 👉 Example: • Staphylococcus → Catalase Positive • Streptococcus → Catalase Negative ⸻ ⚙️ Principle of Catalase Test Toxic substances are formed in our bodies and during bacterial metabolism, such as: • Hydrogen peroxide (H₂O₂) • Superoxide radicals 👉 These substances are harmful to cells. 👉 Therefore, some bacteria possess the catalase enzyme, which detoxifies these toxic substances. 👉 When we expose bacteria to hydrogen peroxide: • If catalase enzyme is present → bubbles will form • If enzyme is absent → bubbles will not form ⸻ 🧪 Materials Required To perform this test, we need: • Glass slide • Inoculating loop or wooden stick • Hydrogen peroxide (3% or 30%) • Bacterial colony ⸻ 🧫 Procedure (How to perform the Catalase Test?) The catalase test is very easy; just follow the steps below: Step 1 Take a clean and dry glass slide Step 2 Pick up a bacterial colony with a sterile loop or stick Step 3 Place the colony on the slide Step 4 Now, add a drop of hydrogen peroxide to it Step 5 Observe the immediate reaction ⸻ 👀 Observation (What to observe?)
