Sterilization is the initial step in tissue culture. The tools and materials to be used must be sterilized first. Sterilization of tissue culture materials occurs after the media is prepared. This is done using a device called an autoclave. When sterilizing media in an autoclave, ensure the temperature is maintained at 121°C for 30 minutes. Excessive sterilization will damage the media and result in the loss of nutrients, which will hinder plant growth. After autoclaving, the bottles are placed in plastic bags sprayed with 70% alcohol to maintain sterility and prevent dust from entering.
