Presented By: Dr. Karen Hogg - Experimental Officer - Imaging and Cytometry (KGH) Speaker Biography: Karen Hogg is an Experimental Officer: Imaging and Cytometry at the University of York, Bioscience Technology Facility. Karen underpins the scientific service within the Imaging and Cytometry Laboratory and takes a lead role in the research, operation and method development of cell sorting and analysis. As such, Karen's expertise is utilized for a wide range of diverse applications both within and external to the Department of Biology. Karen is also a co-organizer and tutor on multiple flow cytometry courses. She obtained her PhD from the School of Biology, University of Leeds, U.K. 1999 and worked in a parasite immunology research group for 4 years before taking up her current role. Karen was awarded the Royal Microscopical Society medal for Flow Cytometry in 2016. Also Presented By: Dr. Peter O'Toole - Director of the Bioscience Technology Facility, Head of Imaging and Cytometry, Vice-President of the Royal Microscopical Society (POT) Speaker Biography: Peter is Director of the Bioscience Technology Facility at the University of York covering the broad technologies required for a multi-disciplined Department of Biology. He has also built up and heads the highly successful Imaging and Cytometry Labs which includes an array of confocal microscopes, flow cytometers, electron microscopes and novel instrumentation. Peter gained his PhD in the Cell Biophysics Laboratory at Essex and has been involved in many aspects of fluorescence imaging and cytometry. Research is currently focused on both technology and method development of novel probes and imaging modalities. His lab provides research support to many academics and commercial organisations and he has ongoing collaborations and consultancy contracts with many leading microscopy and cytometry companies. Peter serves on an array of committees for e.g. BBSRC , Vice-President of the RMS and Steering committee for ELMI. Peter is also heavily involved with microscopy and cytometry training through a variety of international courses. Webinar Abstract: Little and Large - Using the flexibility of the MoFlo Astrios from sorting bacteria, through the more classic blood and human cells through to plant protoplasts! Our MoFlo Astrios serves or multidisciplinary Department of Biology through our open access Bioscience Technology Imaging and Cytometry Core Lab at the University of York. As a core instrument with internal users with very varied demands with the added variety coming in from external users, it is vital that our cell sorter can cope with as diverse a range of samples types as possible. The presentation is not simply about the samples, but how we approach sorting varied sample types to give us the best success rates for each samples type with tips and tricks along the way. We will present data demonstrating the variety of cell sorts that have been undertaken. This will look at bacterial samples that are being selected for their fluorescent protein expression through to the large plant protoplasts. These samples require the range of nozzle sizes and very different pressure tolerances. We also discuss our immunology driven data sets that are being used for downstream RNA and Mass Spectrometry analyses followed by the need for sterile sorting for human prostate research and samples that require multiwall plate sorting and the strategies used to get near 100% success rates for single cell dispositions.
