Basics of Phytochemistry (12) = Introduction and History of Chromatography    • Chromatography  | Introduction and History...   Chromatography is the technique is used primarily for the separation of the components of a sample in which the components are distributed between two phases of which one is stationary (Solid/ liquid) while other is mobile phase (liquid/ gaseous). Chromatographic technique are generally classified on the basis of separation principle of it i.e., adsorption, partition, ion exchange or sieving. These are as Paper, Column, TLC, HPTLC, HPLC, GLC, GSC, Ion exchange chromatography, Gel chromatography etc. All chromatographic techniques are useful for Qualitative as well as Quantitative applications. Gradient and isocratic elution technique preferred to separation on the basis of are generally retention factor (R) or retention volume, area under curve. Developments in the field of chromatography have facilitated parallel rapid advancement in our understanding of complex phytochemical profiles of valuable plant drugs. It has been one of the most useful techniques of general application not only to the field of phytochemical analysis, but also to a host of other biochemical processing sectors requiring rapid and efficient resolution of complex mixtures of chemicals. Based on the physical nature of the mobile and stationary phase: On the basis of physical nature of mobile phase, there are two types of chromatography: 1. Gas chromatography 2. Liquid chromatography On the basis of stationary phase, physical nature there are the following types: 1. Gas-solid chromatography 2. Gas-liquid chromatography 3. Liquid-solid chromatography 4. Liquid-liquid chromatography Based on the mechanism of separation: The method of chromatography uses various types of mechanisms to separate analyte. On the basis of mechanism of separation, it is of following types: 1. Partition chromatography 2. Adsorption chromatography 3. Size exclusion chromatography 4. Affinity chromatography 5. Ion exchange chromatography APPLICATION OF CHROMATOGRAPHY 1. In the pharmaceutical sector: In detection of presence of trace elements or chemicals. 2. This is also used for the separation of compound on the basis molecular weight and composition of elements. 3. In the detection of unknown compound and in the purity of mixtures. 4. In the development of drug. 5. In the Chemical industry: 6. HPLC and GC are very much used for detecting various contaminants such as polychlorinated biphenyl (PCBs) in pesticides and oils. 7. In the Food Industry: In the detection of food spoilage and additives. 8. In the quantitative and qualitative analysis of nutrients in food. 9. In Forensic Science: In forensic science, this technique is used in the analysis of crime scenes like hair and blood samples. 10.
