This time, we invited Dr. Alessandro Loiola, a skeptic about Covid vaccines, to talk with Dr. Luciana Leite, director of the Vaccine Development Laboratory at the Butantan Institute. To help clarify this issue further, we also sought the assistance of a second doctor, who provided us with the following statement: "Regarding the safety of viral vector vaccines, from the preclinical studies onwards, vaccines whose vectors (modified viruses) were not capable of replicating were chosen. A virus consists of a protein base (capsid) that serves to protect the virus's genetic material and aid in cellular infection. What is determinant for the pathological function that the virus performs is its genetic material. More specifically, which proteins will be decoded by it. Typically, among the viral proteins, there are functional proteins (which regulate replication, for example, and which may have a biological function within the host cells) and proteins that will compose the new viral capsids that will allow the new viruses to leave the cells and infect new cells in the host and in new hosts. Precisely to prevent these viral vectors, which serve as carriers of genetic material for vaccines, from replicating, the viruses that gave rise to them are depleted of their original genetic material, becoming practically the capsule. And the new genetic material of interest is introduced into it. In the case of Covid vaccines, a DNA sequence whose transcription product is the S protein of SARS-CoV-2. And it is important to note here that these new “viruses” - the vaccine - are not capable of replicating. Therefore, they will not be transmitted to other people and/or other animals. In science, we always deal with probabilities. That is why, before a drug or vaccine reaches the population, we have a series of very rigorous steps regarding safety during human testing. And before that, there is a whole discussion in ethics and scientific committees about which products and technologies can be used and tested in order to reduce the possibility of accidents or unexpected and unwanted phenomena. Thus, when it comes to a non-replicating viral vector vaccine, the chance of there being any kind of recombinant event of the vector in nature tends to zero. Interestingly, in recent weeks we have seen the rigidity of these rules when our respected health agency, ANVISA, did not approve Sputnik V precisely because of the possible presence of replicating viral vectors. If by On the one hand, the risk of non-replicating viral vector vaccines recombining in nature tends to zero, but we cannot say the same about the risk of recombination of SARS-CoV-2 itself. Letting an epidemic get out of control makes this chance a real and possible possibility. And the best solution for this is to vaccinate the population. Before anyone asks about the time frame for side effects, how long it takes to safely identify possible side effects of a vaccine, the answer is: a short time. The viral antigens contained in the vaccines are metabolized and destroyed in a short time (hours or a few days), and the adaptive immune response that would be capable of an autoimmune reaction takes between 2 and 4 weeks to manifest. Therefore, very rare side effects (such as thromboses linked to the AstraZeneca vaccine) only appear with use in hundreds of thousands of people. And it is not related to a long exposure time. Even an event of this nature (estimated at 1:350,000) is 3 times rarer than a death from anesthesia in surgery.
