Calculation of Pollen Germination Biology – PU 2 Laboratory Experiment Aim - To calculate the percentage of pollen germination in a given pollen sample. Principle - When pollen grains are placed in a suitable medium containing sucrose, boric acid and calcium ions, viable pollen grains germinate by producing a pollen tube. The number of germinated pollen grains is counted and expressed as a percentage of the total pollen grains observed. Procedure (Brief) - Collect fresh pollen grains from a mature anther. Place them on a clean glass slide containing a drop of pollen germination medium. Cover with a coverslip and keep the slide in a moist chamber. Observe the slide under a microscope after a fixed time. Count the total number of pollen grains and the number of germinated pollen grains.
