Improve your resume and qualify for the job market with the best teachers! *** 2.3 Salmonella-Shigella Agar (SS) 2.3.1 Principle SS agar has components (bile salts, brilliant green, and sodium citrate) that inhibit Gram-positive microorganisms. • The incorporation of lactose into the medium allows differentiation between lactose-positive microorganisms (bacteria that ferment lactose produce acid which, in the presence of the neutral red indicator, results in the formation of pink colonies) and bacteria that do not ferment lactose, which form transparent colonies. Sodium thiosulfate and ferric citrate allow the detection of H2S, evidenced by the formation of black colonies in the center. 2.3.2 Usefulness To select and isolate Salmonella and Shigella species in stool, food, and water samples. 2.3.3 Formula/Product Commercial medium: SS Agar. 2.3.4 Procedures Weigh and hydrate the medium according to the manufacturer's instructions. Heat the medium until the agar melts. Do not autoclave – This medium does not need to be sterilized. Cool to 50°C and distribute 20 to 25 mL into sterile 90 mm Petri dishes.  Leave at room temperature until cool; pH: 7.0 +/- 0.2 (25°C). Wrap the plates with transparent PVC plastic and store in a refrigerator at 4 to 8°C. 2.3.5 Quality control Positive: Salmonella typhimurium ATCC 14028. Negative: Staphylococcus aureus ATCC 25923. 2.3.6 Inoculation Inoculate the plates and incubate for 18 to 24 hours. If negative after 24 hours, re-incubate for another 24 hours. 2.3.7 Interpretation Original color of the medium: orange-red. • Colonies with a black center (H²S) or colorless colonies: suspected Salmonella. Colorless colonies: suspected Shigella spp. Pink or red colonies: suspected Escherichia coli or Klebsiella spp. Non-lactose fermenting bacteria are colorless. Lactose fermenting bacteria appear pink. 2.3.8 Storage and shelf life Store packaged at 4 to 8°C for 1 month. 2.3.9 Recommendations Absence of growth or scarce growth, re-incubate the plate for another 24 hours. Do not autoclave, as the high temperature degrades the sugar contained in the medium.
