For Notes Please download App: https://mbpra.courses.store/321242?ut... #MethodsOfImmobilization. there are different methods of immobilization 1) adsorption of Methods of Immobilization 2) covalent bonding of Methods of Immobilization 3) entrapment of Methods of Immobilization 4) encapsulation of Methods of Immobilization 5) crosslinking of Methods of Immobilization 1) adsorption of Methods of Immobilization- is a simplest method in which physical binding of enzyme on the surface of an inert support the support material may be inorganic like alumina, silica gel , calcium phosphate, gel , glass etc. the adsorption of enzyme molecule attach with the weak forces like vonderwall forces, hydrogen bonds. 2) entrapment of Methods of Immobilization- enzymes are entrapped in a support or inside of fibres or polymer membrane. the size of the matrix pores is such that the enzyme is retained while the substrate and the product molecules pass through. matrix is used for and trapping- polyacrylamide gel, collagen , gelatin starch, cellulose , silicone rubber. 3) encapsulation of Methods of Immobilization: it is a type of entrapment in which a spherical particle formation where in a liquid or suspension is enclosed in a semipermeable membrane. membrane may be polymeric, lipoidal , lipoprotein or non ionic in nature. 4) covalent binding of Methods of Immobilization- covalent bonds between the chemical groups of enzyme and the chemical group of the support . the strength of binding is very strong. covalent bond formation with the side chain of amino acid of the enzyme most commonly employed matrixes are agrose, cellulose and polyacrylamide sepharose. 5) crosslinking of Methods of Immobilization- in this solid support is absent and enzyme is linked with multifunctional reagent which act as linker to connect the enzyme molecule in 3D cross-linked manner. these Linking need polyfunctional reagents. several reagents are glutaraldehyde, diazo benzidine , hexamethylene dii isocyanate, toluene dii isocyanate.
