After presenting the learning objectives, such as how to use plate count methods and assess the sanitary quality of analyzed products, she describes the process in detail. It begins with sample preparation, taking 10 grams of the sample and adding the eluent to dilute it properly. The sample is then homogenized using a stomacher-type paddle grinder to obtain a stock suspension. From this, decimal serial dilutions are performed. Once the dilutions are ready, the plate count is performed. It is essential to have sterile Petri dishes, flasks with agar culture medium, and adequate laboratory equipment for working under aseptic conditions. The video shows the process of plating the inoculum in duplicate on the plates using aseptic techniques to avoid contamination. Culture medium is added to each plate and homogenized. Once the agar has solidified, the plates are incubated upside down at 30°C for 72 hours. At the end of the incubation, the colonies on the plates with between 30 and 300 colonies are counted, selecting the appropriate dilution for counting. The average of the duplicate plates is taken and multiplied by the inverse of the dilution factor to obtain the number of colony-forming units per gram or milliliter of sample. This method estimates the total number of microorganisms in a sample and reflects its sanitary quality, but it does not identify the type of microorganism. Despite being a useful indicator of food safety, it has limitations in detecting pathogens or their toxins.
